Immunomodulatory Effects of Multi-Component Dietary Supplementation on Humoral and Cellular Immune Responses: A Randomized Controlled Trial

Abstract

Multi-component dietary supplements targeting immune function represent a growing clinical intervention strategy. This randomized, double-blind, placebo-controlled trial evaluated the immunomodulatory effects of IMMUN-5 — a capsule-based supplement containing tissue hydrolysate (1 mg), ascorbic acid (25 mg), and menthol (5 mg) — in 120 adults with immunodeficiency-associated conditions over 60 days. Participants received IMMUN-5 (two capsules twice daily) or identical placebo. Outcomes included serum IgG, IgM, and IgA concentrations, CD3+/CD4+/CD8+ T-lymphocyte percentages, NK cell frequency and cytotoxicity, and phagocytic index, assessed at baseline, Week 4, and Week 8. The IMMUN-5 group demonstrated significantly greater increases in IgG (+34.7%), IgM (+41.1%), CD3+ T cells (+17.6%), NK cytotoxicity (+42.4%), and phagocytic index (+44.4%) compared with placebo (all p<0.001). The CD4+/CD8+ ratio remained stable. Adverse events were mild and comparable between groups. IMMUN-5 significantly enhanced both humoral and cellular immune parameters without evidence of immunological dysregulation.
